Post Doctoral Associate Med-Ophthalmology - Pennsylvania-Pittsburgh - ( 24000571 ) A postdoctoral position is available in the Paul R (Kip) Kinchington lab of the University of Pittsburgh, Research in Ophthalmology Department at the Vision Institute. The research will focus on the Molecular Biology of the Herpesvirus Varicella Zoster Virus. The specific duties are to conduct research on the molecular determinants of attenuation and latency of the live varicella zoster virus (VZV) vaccine, using tissue culture systems, models of differentiated and organized human skin, and human neuron cultures derived from human embryonic and Induced pluripotent stem cells and neuroprecursor systems. The projects extend to the understanding of how limited viral gene expression during the latent state contributes to latency in and reactivation of VZV from sensory human neuron models. The individual will join a team conducting similar research and the applicant will be encouraged to develop their own personal research programs aligned to the broad goals of our laboratory and our collaborators. As part of the Postdoctoral experience, applicants will receive mentoring for the development of research proposals and in career development activities through the office of academic post-doctoral development at the University of Pittsburgh. Specific duties include: - Eventual supervision and cooperation with other members of the laboratory - Conduction of initially guided methods including viral manipulation by Bacterial Artificial Chromosome (BAC) mutagenesis, use and development of CRSPR-cas9 approaches, - In vitro characterization of recombinant VZV virus in simple and complex cultures (differentiated human skin and corneal epithelia, human skin explants, and human sensory neuron models) - Gene expression studies and their analyses at bulk and single-cell level. - Persons with some bioinformatic skills in genome assembly, RNAseq, and ChiPseq analyses would be at an advantage, as these methods are in development in the laboratory A PhD is required in a relevant field (Microbiology (Virology), Molecular Biology or Cell Biology).
